Yes you do need a hosting connection per region. When you create a connection with the wizard, it will only see the AZs available for the region it has been set to. If you need another region, you'll need a new hosting connection because the wizard won't allow you to pick another region from an existing hosting connection. So you also need at least two Cloud Connectors per region. I can share some screenshots if you need ;)
